A bacterial stimulation assay for bronchoalveolar lavage immune cells from young children with cystic fibrosis

چکیده
Cystic Fibrosis (CF) is primarily a progressive lung disease, characterized by chronic pulmonary infections with opportunistic pathogens. Such typically commence early in life, producing an inflammatory response marked IL-8 chemokine production and neutrophilic infiltration, major contributory factors CF progression. Studying this inflammation, especially critical for developing new strategies preventing or slowing disruption to the structural integrity of airways. However, evaluating immune responses bronchoalveolar lavage (BAL) cells from children faces technical challenges, including contamination carried due pre-existing low cell number availability. Here, we describe technique preparing BAL young using those bacterial stimulation assay. Initial antibiotic treatment proved essential resident bacteria overgrowing cultures, non-specifically activating cells. ACTB, identified as optimal reference gene, was validated accurate analysis gene expression these Pseudomonas aeruginosa Staphylococcus aureus were used stimulants evaluate CF. Addition gentamicin prevented overgrowth, although if added after 3 hours culture extremely variable resulted, causing suppressive effect some cultures. 1 hour completely effect. This then able reproducibly measure S. P. aeruginosa, co-stimulation both bacteria.

Molecular identification of bacteria in bronchoalveolar lavage fluid from children with cystic fibrosis.
Culture of bronchoalveolar lavage fluid (BALF) is the gold standard for detection of pathogens in the lower airways in cystic fibrosis (CF). However, current culture results do not explain all clinical observations in CF, including negative culture results during pulmonary exacerbation and inflammation in the absence of pathogens. We hypothesize that organisms not routinely identified by cultur...

Bronchoalveolar lavage (BAL) performed in specialist centres has improved the understanding of infant cystic fibrosis (CF) lung disease. As most researchers sample from a single lobe, it was determined whether BAL results could be generalized to other lung segments. Thirty-three CF children, aged 1.5-57 months, underwent in random order sequential BAL of their right middle and lingula lobes. Sp...
متن کاملCyanide in bronchoalveolar lavage is not diagnostic for Pseudomonas aeruginosa in children with cystic fibrosis.
Early detection of the cyanobacterium Pseudomonas aeruginosa in the lungs of young children with cystic fibrosis (CF) is considered the key to delaying chronic pulmonary disease. We investigated whether cyanide in bronchoalveolar lavage (BAL) fluid could be used as an early diagnostic biomarker of infection. Cyanide was measured in 226 BAL samples (36 P. aeruginosa infected) obtained from 96 in...

The Standardisation Committee of the European Cystic Fibrosis Society Clinical Trial Network has undertaken the evaluation of clinical end-points for therapeutic interventions regarding their use in multicentre clinical trials in cystic fibrosis (CF). This review of biomarkers in bronchoalveolar lavage (BAL) is part of the group's work. The aims of this project were: 1) to review the literature...
متن کاملHigh concentrations of pepsin in bronchoalveolar lavage fluid from children with cystic fibrosis are associated with high interleukin-8 concentrations.
BACKGROUND Gastro-oesophageal reflux is common in children with cystic fibrosis (CF) and is thought to be associated with pulmonary aspiration of gastric contents. The measurement of pepsin in bronchoalveolar lavage (BAL) fluid has recently been suggested to be a reliable indicator of aspiration. The prevalence of pulmonary aspiration in a group of children with CF was assessed and its associat...
